L-Tryptophan vs 5-HTP: What Is the Difference and When Should You Get Advice?

L-tryptophan and 5-HTP sit on the same biochemical route, but they are not the same supplement. L-tryptophan is an essential amino acid found in protein foods. The body can convert it into 5-hydroxytryptophan, usually shortened to 5-HTP, and then into serotonin. That neat arrow diagram is where many explanations stop. The useful comparison starts with everything the diagram leaves out: dose, competing pathways, study quality and medicine interactions.

If you are comparing L-tryptophan vs 5-HTP for sleep or mood, neither should be treated as a simple serotonin switch. Research is mixed and product doses differ widely. Anyone taking an antidepressant, migraine medicine or another medicine that affects serotonin should ask a pharmacist or prescriber before using either.

The pathway without the sales pitch

Dietary protein supplies L-tryptophan alongside other amino acids. After digestion and absorption, tryptophan has several possible jobs. It can support protein synthesis, enter the kynurenine pathway or contribute to production of niacin. A smaller proportion can be converted to 5-HTP and then serotonin. Serotonin itself does not simply travel from the gut into the brain, so claims that one capsule directly “tops up brain serotonin” oversimplify the biology.

5-HTP is one biochemical step closer to serotonin, but “closer” does not mean clinically better or safer. It changes the interaction question, and the long-term evidence base remains limited. The body regulates pathways; a diagram does not tell you how a person will feel.

What the sleep research can and cannot say

A 2022 systematic review and meta-analysis reported some improvement in one sleep measure after tryptophan supplementation, with findings more apparent in studies using at least 1g a day. The review did not turn every sleep outcome positive, and studies varied. This matters because gram-level research cannot be used to imply that a capsule containing a few hundred milligrams will reproduce the result.

Sleep is also not one outcome. Time to fall asleep, number of awakenings, total duration, next-day alertness and subjective quality are different measurements. A study can move one and not another. Before buying anything, decide what problem you are trying to solve. A racing mind at bedtime, waking with pain, shift work, snoring and restless legs need very different conversations.

Why mood claims need even more restraint

A Cochrane review of tryptophan and 5-HTP for depression found only two trials with 64 participants that met the review's quality criteria. The authors concluded that the evidence was insufficient to guide clinical use. That is not a platform for self-treating low mood or replacing prescribed care.

If low mood is persistent, severe, linked with thoughts of self-harm or affecting daily life, use appropriate NHS and clinical support. Supplements do not diagnose the cause and should not become a reason to postpone help.

The interaction question comes before the ingredient question

Serotonin syndrome is uncommon but potentially serious. It can occur when medicines or substances that increase serotonin are combined. Symptoms can include agitation, confusion, sweating, diarrhoea, muscle twitching, shivering, a rapid heartbeat or a high temperature. Read the NHS medication-safety guidance on serotonin syndrome and seek urgent help for a suspected severe reaction.

Ask a pharmacist before L-tryptophan or 5-HTP if you take antidepressants, tramadol, some migraine medicines, lithium, linezolid, St John's wort or any product described as serotonergic. Bring the actual packets or a medicines list. “Natural” does not answer an interaction question.

  4. What else is in the formula? Count duplicate B6, magnesium and niacin from other products.
  5. What would make you stop? New symptoms, no useful change after a defined period, or a clinician advising against it.

Use a symptom diary before changing the pathway

For seven days, record bedtime, estimated time to fall asleep, awakenings, wake time, caffeine after lunch, alcohol, exercise and medicines. If mood is the concern, record when it drops, how long it lasts and what it stops you doing. The diary is not a diagnosis. It helps separate a vague wish to “boost serotonin” from a specific problem that can be discussed properly.

If sleep timing shifts from day to day, stabilising wake time and morning light may be a cleaner first experiment. If worry keeps you awake, write the next action before bed rather than adding several calming ingredients. If pain, snoring or restless legs interrupts sleep, the route is different again.

Only change one supplement variable at a time. Combining tryptophan, 5-HTP, magnesium, herbs and a sleep drink creates an impossible experiment and a larger interaction surface. A modest, measured routine gives you a better answer than an impressive stack.

Do not shop from the pathway alone

The pathway diagram explains why researchers are interested. It cannot select a supplement for you. Food provides tryptophan within protein, while sleep and mood problems have many causes. A product becomes a reasonable option only after the question, medicine check, dose and evidence line up. If one of those pieces is missing, the honest answer is to pause.
